  when I change a new locale can affect all old data exists in spreadsheet.
  
  ex A1: have format DateTime en-US. I change locale to en-UK. A1 can auto change to DateTime en-UK, right?
  
  or i will setting format DateTime cell in A1 by manual

  Yes, once you change the culture all the existing values will be updated according to the new culture.
